Benzodiazepine Pharmacology

One receptor-level distinction organizes benzodiazepine prescribing: these drugs amplify GABA-A signalling rather than independently activating the channel. That helps explain their comparatively wider overdose margin when taken alone—and why the margin can disappear with opioids, alcohol, or other central nervous system depressants. For an NP, the pharmacology becomes a set of linked decisions: match the agent, route, and duration to the indication; account for age, hepatic function, respiratory vulnerability, and interaction burden; identify dependence before stopping therapy; and manage airway or seizure emergencies without reflexively reaching for reversal.
